Combining CSS and JS can help reducing the request count, but you should make sure not to use it or exclude some CSS/JS files if the size of the combined file turns to be too big as that could have a negative effect.
You can enable Combine CSS and JS by going to SpeedyCache --> File optimization in the WordPress admin, as shown in the screenshot below.
